New gene assocaited with androgenetic alopecia found

A new gene called dickkopf 1 (DKK-1)has been found to cause DHT driven balding. Its levels are found to be increased in persons with bald scalp.
In vitro studies have shown that neutralizing this gene reverses DHT-induced epithelial cell death in cultured hair follicles.

This info is over a year old.

DKK-1, a short name for it, is upstream from apoptosis of keratinocytes. Therefore some of the dead keratinocyte cells that might be in the infidulum, where the first inflammation is seen in AGA, might have died due to DKK-1 upreged in the dermal papilla. Perhaps this is the genesis of the immuno response or so it has been speculated.

I like this bit. Now only if that was possible in vivo and trials have passed and problem solved.

Dihydrotestosterone-Inducible Dickkopf 1 from Balding Dermal Papilla Cells Causes Apoptosis in Follicular Keratinocytes

Recent studies suggest that androgen-driven alteration to the autocrine and paracrine factors produced by scalp dermal papilla (DP) cells may be a key to androgen-potentiated balding. Here, we screened dihydrotestosterone (DHT)-regulated genes in balding DP cells and found that dickkopf 1 (DKK-1) is one of the most upregulated genes. DKK-1 messenger RNA is upregulated in 3–6 hours after 50–100 nM DHT treatment and ELISA showed that DKK-1 is secreted from DP cells in response to DHT. A co-culture system using outer root sheath (ORS) keratinocytes and DP cells showed that DHT inhibits the growth of ORS cells, and neutralizing antibody against DKK-1 significantly reversed the growth inhibition of ORS cells. Analysis of co-cultured ORS cells showed a significant increment of sub-G1 apoptotic cells in response to DHT. Also, recombinant human DKK-1 inhibited the growth of ORS cells and triggered apoptotic cell death. In addition, DHT-induced epithelial cell death in cultured hair follicles was reversed by neutralizing DKK-1 antibody. Moreover, immunoblotting showed that the DKK-1 level is up in the bald scalp compared with the haired scalp of patients with androgenetic alopecia. Altogether, our data strongly suggest that DHT-inducible DKK-1 is involved in DHT-driven balding.

Researchers are always giving rather dubious names to proteins as you can see by looking at many charts of signal transduction. also close to the world of hair and hairloss the Hedgehog family of proteins that some researchers decided to call the member they discovered “Sonic”. Some doctors have objected to this as they say telling a parent “Your child is dying because they have a defective gene for Sonic Hedgehog” is not something they look forward to.

German scholars believe the translation has been lost over thousands and thousands of years…so I’ll agree to disagree. :slight_smile:

actually this is much older than one year. i have read about it a few years ago (maybe 3?). it is correlated with some BMPs and (therefore) with the wnt pathway but I have forgotten the exact mechanism and I am sure much knowledge has been added to this.

Im from German…Dickkopf Dick=fat Kopf=head
But the right translat is,if somone to persist…we than say…you are a DICKKOPF;-)

Man hat mir gesagt,mein Englisch ist gut genug,und das man mich einwandfrei versteht.
Bist du auch bei Alopezie.de.Dein Englisch ist deutlich besser als meins,vielleicht kannst ja dort mal im Forschungsforum bissel was schreiben.Mir fällt es schwer bei diesen ganzen Müll hier wertvolle Infos rauszulesen.Wenn es hier nix neues gibt,fangen sie an Artikel von vor drei Jahren rauszuholen:-(

jo man versteht dich ja auch also alles kein problem.
ich hab früher mal in dem forum geschrieben, aber das ist sinn- und zwecklos. da kommt nix bei rum. ich beschränk mich schon seit geraumer zeit aufs lesen und ganz selten auch mal was schreiben.
